AIM: Umbilical cord blood plasma contain higherhematopoietic stimulatory activities than adult peripheral blood plasma. IL-3 is regardedas multilineage hematopoietic growth factor that acts on primitive pluripotential stemcells and progenitor cells of every lineage except T and B-lymphoid lineage. GM-CSFregulate proliferation and differentiation of multilineage progenitors, myeloid, erythroid, megakaryocyte precursors. This experiment was designed to investigate whetherIL-3 and GM-CSF contribute to hematopoietic stimulating activities of cord blood plasma.METHODS: IL-3 and GM-CSF levels of cord blood plasma levels were assessed in 33 termneonates by enzyme-amplified sensitivity immunoassay (ELISA), and 9 health adultperipheral blood plasma were assessed as controls. RESULTS: Plasma median levels of IL-3in cord blood and adult blood were 5 ng/L vs. 5 ng/L, and ranges were 0~35 ng/L vs. 0~28 ng/L. There was no statistical significant difference between two groups (P>0.05). GM-CSF was detected in only 12% cord blood (median 19.5 ng/L, range 8~44 ng/L), while GM-CSF levels were undetectable in all adult samples, but difference in GM-CSF levelsbetween two groups did no reach statistical significance (P>0.05). CONCLUSION: The IL-3and GM-CSF of cord plasma may play minor rolses in stimulating the ex vivo hematopoiesis.The hematopoietic stimulatory activities of cord blood plasma require synergisticalactions of other hematopoietic growth factors.
